异尖科线虫诱导长江刀鲚免疫反应研究

摘要:
为探究长江刀鲚TH1细胞、MIF、IL-10、TNF-α、IgM、IgT和IgD的常规水平及异尖科线虫寄生所导致的免疫变化,采用ELISA法分别测定了空白组和3个寄生组的脾脏、头肾和血清中7种免疫分子的表达水平。结果显示:(1)空白组脾脏、头肾和血清中免疫分子水平均表现为IgD(P<0.01)> IgM(P<0.001)>IgT(P<0.001)> MIF(P<0.001)> TH1细胞(P>0.05)>TNF-α(P>0.05)>IL-10,且血清中免疫分子水平显著高于脾脏和头肾(P < 0.001);(2)各寄生组脾脏中7种免疫分子水平均显著高于空白组(P<0.001),头肾中除IgM外的6种免疫分子水平均显著高于空白组(P<0.05),血清中仅IL-10水平显著高于空白组(P<0.001);(3)整体而言,长江刀鲚寄生组IL-10水平随着寄生数量增加而下降,TH1细胞随着寄生数量的增加呈下降后上升趋势,MIF、TNF-α、IgD、IgM和IgT则与TH1细胞相反,且脾脏(IgT、TNF-α、TH1细胞、MIF、IgM)、血清(TH1细胞、IgD、IL-10、TNF-α)和头肾(IgT、TNF-α)中免疫分子水平与寄生虫数量间的相关关系均达到显著水平。综上,异尖科线虫寄生可增加长江刀鲚免疫分子的表达水平,诱导炎症反应和适应性免疫反应的发生,脾脏、头肾和血清中部分免疫分子水平与异尖科线虫数量间的相关性显著,其中血清TH1细胞、脾脏中IgT和TNF-α可作为长江刀鲚应对异尖科线虫寄生的免疫功能测定优选指标。

Abstract:
To investigate the level of seven immune molecules (TH1 cells, MIF, IL-10, TNF-α, IgM, IgT and IgD) in Coilia nasus influenced by Anisakidae-infected, the expression of seven immune molecules in the spleen, head kidney and serum were measured by ELISA. The results showed that: (1) For the blank group, the level of immune molecules in spleen, head kidney and serum all showed the trend: IgD (P < 0.01)>IgM (P < 0.001)>IgT (P < 0.001)>MIF (P < 0.001)>TH1 cell (P > 0.05)>TNF-α (P > 0.05)> IL-10, and the expression of seven immune molecules in serum were significantly higher than those in spleen and head kidney (P < 0.001). (2) For parasitic group, the level of seven immune molecules in spleen of C. nasus were significantly higher than those of the blank group (P < 0.001). The level of six immune molecules except IgM in head kidney were significantly higher than those in the blank group (P < 0.05), and the level of IL-10 in the serum of head kidney was significantly higher than the blank group (P < 0.001). (3) Overall, the IL-10 level of parasitic group in C. nasus decreased with the increase of parasitic number. The TH1 cell level of parasitic group in C. nasus decreased firstly but then increased with the parasitic number increasing, while MIF, TNF-α, IgD, IgM and IgT showed the opposite tendency.
