一株耐盐菌的筛选、鉴定及对苯酚和镉耐性特征

摘要:
为了研究高盐环境下菌株生长情况及对污染物的耐性特征,从危险废物填埋场渗滤液中筛选得到一株耐盐菌,编号为WS-1。通过对其形态进行观察,以及革兰氏染色和16S rDNA基因序列分析,可以鉴定该菌株为葡萄球菌属(Staphylococcus sp.)。通过调节培养基的盐度、pH,研究其对WS-1生长的影响,并考察WS-1对苯酚和镉的耐受性。确定WS-1在盐质量分数为5%~10%、pH为7.0、温度30℃时生长最好。WS-1在苯酚浓度0~1 000 mg·L-1的范围内均可很好生长,低浓度苯酚能促进菌株生长,高浓度苯酚会对菌株生长产生抑制作用,72 h内,菌株对苯酚的降解率可达38%左右。在Cd2+浓度在1~5 mg·L-1时,WS-1生长不受影响,当Cd2+浓度达到10 mg·L-1,抑制作用明显,延长WS-1生长周期。低浓度时WS-1对Cd2+去除率在90%左右,高浓度Cd2+去除率在40%左右,效果不明显。WS-1可为高盐度含有机废物和重金属废水生物处理提供菌株资源。

Screening, identification and tolerance to phenol and cadmium of a salt-tolerant strain

Abstract:
In order to study the growth of a strain and its tolerance to pollutants in high salt environment, a salt-tolerant strain was screened from a hazardous waste landfill leachate, numbered WS-1. By observing its morphology, as well as Gram staining and 16S rDNA gene sequence analysis, the strain was identified as Staphylococcus sp. Effects of pH and salinity on the growth and the tolerance to phenol and cadmium of strain WS-1 were investigated, the results showed that the optimum conditions of WS-1 were salinity 5%~10%, pH 7.0, 30°C.The strain WS-1 could grow well in the range of phenol concentration 0-1000 mg·L-1, which growth was promoted at low- concentration and inhibited at high-concentration of phenol. The degradation rate of phenol by WS-1 was at about 38% in 72 h.When the concentration of Cd2+ was 1~5 mg·L-1, the growth of WS-1 was not affected. However, at high concentration of Cd2+ (10 mg·L-1), the inhibitory effect was obvious and the growth cycle was prolonged of WS-1. At low concentration, the removal rate of Cd2+ was about 40%, at high concentration, the removal rate of Cd2+ was about 40% which not obvious. So, WS-1 shows the potential of application in biological treatment of high salinity wastewater including organic and heavy metal pollutions.
