A型产气荚膜梭菌青海分离株virR基因序列与蛋白结构分析

摘要:
为了探究产气荚膜梭菌virR基因功能及其蛋白结构特点,提取了产气荚膜梭菌青海分离株的基因组,设计引物对virR基因进行PCR扩增和测序,使用生物软件对该基因进行序列分析和蛋白预测。结果显示,产气荚膜梭菌分离株virR基因长为759 bp,其编码252个氨基酸;分离株virR基因与参考菌株SM101、13、ATCC 13124、CP15、Del1、EHE-NE18、FORC 003、FORC 025、JIR4025、JP55、JP838、LLY N11、NCTC2837以及NCTC13170的virR基因核苷酸序列同源性和氨基酸同源性均在97%以上;二级结构预测显示分离株virR蛋白主要由α螺旋结构和β折叠组成;三级结构预测表明分离株virR蛋白有5种可信度较高的结构,蛋白功能位点预测表明分离株virR蛋白具有1个N-糖基化位点、1个cAMP和cGMP依赖性蛋白激酶磷酸化位点、2个蛋白激酶C磷酸化位点、3个酪蛋白激酶Ⅱ磷酸化位点以及1个N-豆蔻酰化位点。

Sequence and protein structure analysis of virR gene of Clostridium perfringens type a isolated strain from Qinghai

Abstract:
In order to explore the function of virR gene and its protein structure in Clostridium perfringens, this study extracted the genome of Clostridium perfringens Qinghai isolated strain, designed primers to PCR amplification and sequencing of virR gene, using DNA Star software and Online server for sequence analysis and protein prediction of this gene. The results showed that the virR gene of Clostridium perfringens isolated strain was 759 bp, which encoded 252 amino acids; the virR gene of isolated strain compared with reference strains SM101, 13, ATCC 13124, CP15, Del1, EHE-NE18, FORC 003, FORC 025,JIR4025, JP55, JP838, LLY N11, NCTC2837 and NCTC13170, the nucleotide sequence homology and amino acid homology of virR gene were all above 97%; The secondary structure prediction showed that the virR protein of the isolated strain was mainly composed of α-helical structure and β-sheets; tertiary structure prediction showed that the virR protein of the isolate has five highly reliable structures, and the functional site prediction indicates that the virR protein of isolated strain has one N-glycosylation site, one cAMP and cGMP dependence, a protein kinase phosphorylation site, two protein kinase C phosphorylation sites, three casein kinase II phosphorylation sites, and an N-myristoylation site.
