不同日龄皖南三黄鸡肌肉中MyoD基因表达及相关分析

摘要:
为研究生肌因子MyoD基因在皖南三黄鸡不同发育阶段肌肉组织中的表达水平及其与肌肉重量的相关分析。分别于胚胎发育的11胚龄、1日龄、70日龄、98日龄采集胸肌和腿肌组织,并测定1日龄、70日龄、98日龄和126日龄的胸肌重、腿肌重和体重,采用实时荧光定量PCR方法分析MyoD基因在不同阶段胸肌和腿肌中的相对表达量。结果表明,胸肌中MyoD基因随着日龄的增加其相对表达量呈降低的趋势;腿肌中MyoD基因随着日龄的增长其表达量呈先增加后降低的趋势。相关性分析表明,MyoD基因的相对表达量与胸肌重呈显著负相关。研究结果提示MyoD基因相对表达量与肌肉发育相关,为进一步研究MyoD基因对鸡肌肉发育的调控作用提供重要的参考。

Expression and correlation of MyoD in muscle tissues at different ages in Wannan Yellow chickens

Abstract:
The aim of this study was to investigate the expression level of MyoD (Myogenic determing factor) gene in muscle tissue at different developmental stages of Wannan Yellow chicken and its correlation with muscle weight. The pectorals and legs muscle tissues were collected at the embryonic age of 11, 1, 70 and 98 days, respectively, and pectorals muscle weight, legs muscle weight and body weight were measured at 1, 70, 98 and 126 days of age. Real-time fluorescence quantitative PCR was used to analyze the expression level of MyoD gene in different stages of Pectorals and legs muscle. The results showed that the relative expression of MyoD gene in the pectoral muscle decreased with the increase of age. The expression of MyoD gene in the leg muscle increased first and then decreased with the increase of age. The result suggests that the relative expression of MyoD gene is related to muscle development, which provides an important reference for further study on the regulation of MyoD gene on chicken muscle development.
