氟苯尼考对肉鸡空肠黏膜的完整性 及免疫相关基因表达的影响

摘要:
研究空肠免疫相关指标及绒毛结构的变化在氟苯尼考致肉鸡腹泻中的作用。40羽1日龄肉鸡适应性饲养7 d后,随机分为对照组、氟苯尼考组(饮水添加100 mg·(kg?d)-1氟苯尼考至35日龄),35日龄检测体重、脾脏与胸腺指数,取空肠组织以HE染色,观察组织切片并测量绒毛长度、隐窝深度且计算绒隐比,荧光定量PCR检测空肠TLR4、MyD88、NF-κB和TNF-α基因表达量。结果表明,与对照组相比,氟苯尼考组肉鸡空肠绒毛长度、绒隐比极显著下降(P<0.01),隐窝深度极显著升高(P<0.01);体重、脾脏指数与胸腺指数极显著下降(P<0.01);TLR4、MyD88、NF-κB与TNF-α mRNA相对表达量极显著升高(P<0.01)。空肠绒毛结构功能改变及免疫相关指标异常表达参与了氟苯尼考致肉鸡腹泻的发生发展。

Effects of flufenicol on integrity of jejunum mucosa and expression of immune-associated genes in broilers

Abstract:
To study the effect of jejunal immune related indexes and villus structure changes in flufenicol-induced diarrhea in broilers.After 7 days of adaptive feeding, a total of 40 1-day-old broilers were randomly divided into control group, flunfenicol group (drinking water supplemented with 100 mg·(kg?d)-1 flunfenicol to 35 days of age). At 35 days of age,the weight,spleen and thymus index were detected,jejunal tissue was stained with HE and then were observed,the length of villus and the depth of crypt were measured and the ratio of villus to crypt was calculated.The relative expression of TLR4,MyD88,NF-κB and TNF-α in the jejunum were detected by real-time PCR.The results showed that compared with the control group,the flufenicol group's villus length and crypt ratio of jejunum extremely significant decreased(P<0.01),the crypt depth of jejunum extremely significant increased (P<0.01);the weight,spleen index and thymus index extremely significant decreased (P<0.01);the relative expression of TLR4,MyD88,NF-κB and TNF-α mRNA were extremely significant increased (P<0.01).The structural and functional changes of jejunal villi and the abnormal expression of immune-related indexes were involved in the occurrence and development of flufenicol-induced diarrhea in broilers.
