响应面法优化酶解鹿角冒渣制备抗氧化活性肽工艺

摘要:
以游离氨基酸含量和抗氧化活性为试验指标,分别采用5种蛋白酶酶解制备鹿角冒抗氧化多肽,对鹿角冒渣为原料制备鹿角冒抗氧化活性肽的工艺进行了研究。结果表明,碱性蛋白酶产物的游离氨基酸含量和抗氧化活性均较高,因此水解用酶选择碱性蛋白酶。通过单因素和响应面回归试验对酶解工艺参数进行优化,得出最佳酶解条件:加酶量6%、底物浓度15%、pH8.5、温度53℃、反应时间4 h,制备的鹿角冒渣抗氧化肽对DPPH·的清除率预测值为74%,验证试验所得实测平均值为(74.65±1.24)%,实测结果与预测值符合良好。

Preparation of antioxidant peptides from antler base residue through enzymatic hydrolysis

Abstract:
This study aimed to prepare antioxidant peptides from antler base residue by enzymatic hydrolysis. Five proteases were used, and amino acid concentration and DPPH radical scavenging activity were evaluated. Results showed that the hydrolysate by alkaline protease possessed the strongest DPPH radical scavenging activity and amino acid concentration. The response surface regression experiment was carried out based on single factor tests, and the optimal parameters for enzymatic hydrolysis were: enzyme concentration, 6%; substrate concentration, 15%; hydrolysis temperature, 53℃; reaction time,4 h; and pH value of 8.5. Under these conditions, the DPPH· scavenging activity of antler base peptides was 74%, and the actual average value was (74.65±1.24)% by verification tests. The measured result was in good agreement with forecast result.
