肉鸭PRKAG2基因表达水平与饲料效率性状的相关性分析

摘要:
为了探讨PRKAG2基因在不同饲料效率肉鸭中的表达规律及与饲料效率性状的相关性,选取具有系谱记录的1 000只H系公鸭,测定21~42日龄体增重和采食量,并计算剩余采食量(RFI)。根据RFI,挑选高低RFI个体各8只,采集胸大肌、腿肌和肝脏,采用qPCR检测高低RFI组肉鸭PRKAG2表达量。结果表明,PRKAG2基因在低RFI组胸大肌的表达量极显著高于高RFI组(P<0.01),低RFI组腿肌中的表达量显著高于高RFI组(P<0.05)。相关性分析表明,PRKAG2在胸大肌中表达量与日采食量、饲料转化率和RFI呈极显著负相关(P<0.01),与代谢体重呈显著负相关(P<0.05);腿肌PRKAG2表达量与日采食量、饲料转化率和RFI呈显著负相关(P<0.05)。

Association of PRKAG2 mRNA expression with feed efficiency traits in meat-type ducks

Abstract:
The objective of this study was to investigate the expression profile of PRKAG2 gene and its correlation with feed efficiency traits in meat-type ducks with high and low feed efficiency. A total of 1 000 pedigreed male ducks were chosen to record body weight gain and feed intake from 21 to 42 days of age, and then residual feed intake (RFI) in the interval was calculated. Based on residual feed intake, sixteen individuals with high and low RFI (8 HRFI vs. 8 LRFI) were selected, and the pectoralis major muscle, leg muscle and liver were taken to isolate total RNA. The relative expression of PRKAG2 between HRFI and LRFI ducks were detected using quantitative PCR. The results showed that the expression level of PRKAG2 in pectoralis major muscle in LRFI were significantly higher than that in HRFI (P<0.01), and its relative expression in leg muscle in LRFI was higher than that in HRFI (P<0.05). The correlation analysis revealed that the expression of PRKAG2 in pectoralis major muscle was significantly negatively correlated with daily feed intake, feed conversion ratio (FCR) and RFI (P<0.01) and significantly negatively related to metabolic body weight (P<0.05). Additionally, the expression of PRKAG2 in leg muscle was significantly negatively related to daily feed intake, FCR and RFI (P<0.05).
