不同有机添加物及柠檬酸钠组合对春兰根状茎增殖的影响

摘要:
通过添加不同浓度猕猴桃汁、香蕉汁、梨汁、苹果汁、白萝卜汁、胡萝卜汁、土豆汁、红薯汁等8种有机物及柠檬酸钠组合,研究其对春兰根状茎增殖及抗褐化效果的影响。结果表明,单独添加猕猴桃汁100.0 g·L -1、梨汁100.0 g·L -1或柠檬酸钠1.0 g·L-1比对照能显著促进根状茎增殖,增殖中生长倍数分别为3.9、2.4和2.2,梨汁和猕猴桃汁处理中,根状茎中细胞分裂相关基因CgCkiCgCKSCgClinsCgE2FCgRb表达量显著提高。1/2MS+NAA 5.0 mg·L -1+活性炭1.0 g·L -1+猕猴桃汁100.0 g·L -1+柠檬酸钠1.0 g·L -1组合对春兰根状茎增殖及抗褐化效果最好,根状茎增殖的生长倍数为4.2,细胞分裂相关基因CgCDKSCgCkiCgCKSCgClinsCgE2FCgRb表达显著提高。因此,在春兰组培快繁过程中猕猴桃汁+柠檬酸钠组合可以有效地促进根状茎增殖,并防止根状茎褐化。

Effects of different organic additives and sodium citrate combination on rhizomes proliferation of Cymbidium goeringii

Abstract:
Rhizomes of Cymbidium goeringii were treated with a variety of organic additives at different concentrations, including kiwifruit, banana, pear, apple, white radish, carrot, potato, sweet potato juice, and a combination with sodium citrate to assess the effects of these agents on rhizome proliferation. The results showed that the proliferation of Cymbidium goeringii rhizomes significantly increased when adding kiwifruit juice (100.0 g·L-1), or pear juice (100.0 g·L-1), or sodium citrate compared with the control and the growth rate reached to 3.9, 2.4 and 2.2, respectively. The expression of cell division related genes (CgCki, CgCKS, CgClins, CgE2F and CgRb) was significantly higher in kiwifruit juice and pear juice treatments compared with the control. The highest growth rate of the rhizome was 4.2 when cultured on 1/2 MS medium containing NAA 9.0 mg·L-1+activated charcoal 1.0 g·L-1 + kiwifruit juice 100.0 g·L-1 + sodium citrate 1.0 g·L-1 with decreased browning and higher expression level of cell division related genes CgCDKS, CgCki, CgCKS, CgClins, CgE2F and CgRb. The results demonstrated that combination of kiwifruit juice+sodium citrate can effectively promote rhizome proliferation and prevent browning of the rhizome in micropropagation of Cymbidium goeringii.
