BMP15基因在不同品种鸡卵泡中的表达规律

摘要:
以淮南麻黄鸡和邵伯鸡为试验素材,研究骨形态发生蛋白15(bone morphogenetic protein 15,BMP15)基因在2个群体卵泡颗粒层和膜层中的表达模式及差异。结果表明,在颗粒层中,BMP15在2个群体的卵泡不同发育时期表达模式均呈先增加后降低的趋势,发育到小黄卵泡(small yellow follicle,SFY)时期呈现表达高峰,显著高于其他各等级卵泡的表达量(P<0.05,P<0.01),之后表达量显著下降;不同时期同一等级卵泡颗粒层中的表达均表现为24周高于43周,小黄卵泡时期表现为24周显著高于43周(P<0.05)。在膜层中,BMP15在2个品种的卵泡不同发育时期表达模式均呈“低→高→低”的趋势,表达高峰出现在小黄卵泡(SYF)时期,显著高于其他各等级卵泡的表达量(P<0.05)。不同时期同一等级卵泡膜层中的表达基本相似,均表现为24周表达量高于43周,在小黄卵泡(SYF)时期膜层表达量表现为24周显著高于43周(P<0.05)。BMP15基因的表达量在各级卵泡的颗粒层和膜层在2个群体间差异不显著。综合BMP15在2个群体不同时期卵泡的颗粒层和膜层中的表达模式研究表明,BMP15在鸡的卵泡发育过程中的表达存在时间、品种和组织差异性。

The expression pattern of the BMP15 gene in follicle of different chicken breeds

Abstract:
The expression pattern of BMP15 in the granular layer and membranous layer during follicle development in Huainan partridge and shaobo chicken was studied. The result showed that in granular layer, the expression pattern of BMP15 in two varieties of chicken’s follicle was first increased and then decreased. The expression reached the peak at small yellow follicle and then significantly decreased (P<0.05, P<0.01). In different periods, the expression of BMP15 in the granular layer of the same grade at 24 weeks was higher than that at 43 weeks, and at small yellow follicle, the expression at 24-week was significantly higher than that at 43-week (P<0.05). In membranous layer, the expression of the BMP15 gene was similar to the “∧”shape in two chicken breeds The expression reached the peak at the small yellow follicle, which was significantly higher than that of other grades (P<0.05). The expression in the membranous layer in the same grade at 24 weeks was higher than that at 43-week. At small yellow follicle, the expression at 24-week was significantly higher than that at 43-week (P<0.05). The BMP15 gene in the granular layer and membranous layer of all grades follicle of Huainan partridge was higher than that of Shaobo chicken with no significant difference. The results suggested that the expression level of the BMP15 gene in follicle showed variations among different development stages, breeds and tissues and small yellow follicle was an important period for follicular development.
