毛竹PheDof4-1基因克隆及表达分析

摘要:
Dof(DNA binding with one finger)蛋白是植物特有的一类转录因子,由多基因共同编码,参与植物转录调控、生长发育及胁迫响应等生物学过程。以毛竹为研究对象,克隆了1个 Dof基因,命名为 PheDof4-1,基因长度为 1 473 bp,推测其蛋白质产物的分子量是 53.2 kDa,等电点为 8.05。qRT-PCR 结果显示,PheDof4-1 在低温和 250 mmol·L-1 NaCl 处理的幼茎中诱导表达,而在 20% PEG8000 处理的根中表达则受到强烈抑制,在叶片中不同处理条件下呈现不同的表达趋势。研究结果为 PheDof4-1 在非生物胁迫中的作用提供理论依据,为竹子分子育种提供参考。

Molecular cloning and expression analysis of the PheDof4-1 gene in moso bamboo (Phyllostachys edulis)

Abstract:
Dof (DNA binding with one finger) proteins forming an important transcriptional factor family encoding by multiple genes are involved in gene transcriptional regulation, development and stress-responses in plants. In this study, one of Dof genes, PheDof4-1 (Gene ID No. PH01000209G1040) was isolated from moso bamboo and the sequence was 1 473 nt in length. The predicted molecular weight of PheDof4-1 protein was 53.2 kDa, and the pI value was 8.05. The qRT-PCR analysis displayed that the PheDof4-1 transcription level in young stem was induced?following exposure to cold stress and salt treatment, however, the expression was restrained in the root under drought treatment. PheDof4-1 showed different expression patterns in the leaf under different stress treatments.
