黄果槲寄生果实多糖对急性酒精肝损伤小鼠的保护作用

摘要:
探讨黄果槲寄生果实多糖(VCFP)对急性酒精性肝细胞损伤小鼠的保护作用。通过一次性高浓度酒精灌胃建立小鼠急性酒精性肝损伤模型,之后将KM小鼠随机分为VCFP低、中、高剂量组和酒精模型组,同时设正常对照组。VCFP组连续灌胃多糖28 d,酒精模型组及正常对照组灌胃等量生理盐水,动态观察生长情况和肝组织形态学结构的变化。结果显示,小鼠酒精灌胃后,模型组谷丙转氨酶(ALT)、谷草转氨酶(AST)、总胆固醇(TC)、三酰甘油(TG)水平、丙二醛(MDA)含量、CAT及GSH-Px活力与正常对照组相比显著升高(P<0.01),体重、肝脏指数和SOD活力显著(P<0.01)降低;VCFP组连续灌胃多糖28 d后,均能显著提高小鼠体重、肝脏指数和SOD活力,降低ALT、AST、TC、TG水平、MDA含量、CAT及GSH-Px活力,肝组织逐渐得到明显修复。研究表明,黄果槲寄生果实多糖可通过提高抗氧化酶活性,清除氧自由基减轻膜脂质过氧化反应,对肝细胞起到保护作用。

The protection mechanism of Viscum coloratum (Kom.) Nakai f.lutescens kitag fruit polysaccharides on acute alcoholic liver injury in mice

Abstract:
To investigate the protective mechanism of V. coloratum fruit polysaccharides (VCFP) on acute alcoholic liver injury in mice, an acute liver injury model was established by one time irrigation stomac. The mice were randomly divided into VCFP-treated groups (low-, medium-, and high-dosage), alcohol model group and normal control group at the same time. VCFP-treated group was given polysaccharide on continuous 28 days by gavage; the alcohol model group and normal control group were injected with the same amount of normal saline, and then the general situation and liver tissue morphological structure changes were observed. The results showed that the levels of ALT, AST, TC, TG, MDA content, and CAT and GSH-Px activities of mice in model group were significantly increased (P<0.01), while the activity of SOD and weight and liver index were lower (P<0.01) compared with the normal control group. After continuously given polysaccharide for 28 days by gavage, the VCFP significantly increased the weight, liver index and SOD activity, while significantly decreased the contents of ALT, ST, TC, TG, MDA and CAT and GSH-Px activities. These results suggested that the VCFP can improve antioxidant enzyme activity and remove oxygen free radicals reducing membrane lipid peroxidation reaction; therefore protect liver cells.
