安徽省长丰县草莓根腐病病原的鉴定

摘要:
对安徽省长丰县随机采集的20个草莓根腐病病样进行病原鉴定,结果分离获得12个菌株。根据培养特征和形态学鉴定,确定分离到的菌株均为尖孢镰刀菌(Fusarium oxysporum);选取真菌通用引物ITS1/ITS4对菌株基因组DNA进行PCR扩增并进行克隆和测序,通过与NCBI数据库序列比对,分子鉴定结果与形态学一致。将分离到的菌株回接到健康草莓幼苗上,2~3周后植株表现出与田间发病相似的症状,重新分离能够得到相同的病原物。基于形态学、分子生物学鉴定及致病性检测,最终确定安徽省长丰县草莓根腐病的病原菌为Fusarium oxysporum

Pathogen diagnosis of strawberry root rot caused by Fusarium in Changfeng county, Anhui Province

Abstract:
Strawberry root rot was observed in Changfeng county, Anhui Province. Twelve strains were isolated from the root of 20 randomly-collected infected plants. The isolates were primarily identified as Fusarium based on their morphological and colony characteristics, and further confirmed as Fusarium oxysporum by analyzing the rDNA-ITS sequence of isolates and morphological characteristics.The healthy strawberries inoculated with the isolate showed the similar symptoms in two to three weeks after inoculation. Fusarium oxysporum was re-isolated from the inoculated pathogenetic strawberry roots. Based on the morphological characteristics, pathogenicity test and molecular analysis, strawberry root rot pathogens from Changfeng county were identified as Fusarium oxysporum.
