核盘菌磷酸泛酰巯基乙胺基转移酶编码基因的 预测与生物信息学分析

摘要:
为了解磷酸泛酰巯基乙胺基转移酶(PPTases)在核盘菌生理代谢与致病过程中的作用,在核盘菌全基因组中寻找PPTases同源物。利用同源比对的方法,预测出核盘菌中PPTases基因,进而分析其编码蛋白的基本理化性质、疏水性、跨膜区、信号肽、亚细胞定位、二级、三级结构和保守结构域,并对PPTases编码氨基酸进行遗传进化分析。结果表明,在核盘菌全基因组中预测存在3个PPTases基因同源物,分别命名为:Ss-Ppt1、Ss-Ppt2和Ss-Ppt3。3个蛋白均为亲水性的非分泌型蛋白,在其二级和三级结构中α螺旋结构和无规则卷曲占主要部分,且三者都含有PPTases的保守结构域SCOP和(或)ACPS。

Prediction and bioinformatics analysis of phosphopantetheinyl transferase gene in Sclerotinia sclerotiorum

Abstract:
Phosphopantetheinyl transferase (PPTase) is an important class protein superfamily existing in eukaryotes. To decipher the function of PPTases in the biological metabolism and pathogenicity of Sclerotinia sclerotiorum, the characters of PPTases genes and the encoded protein were predicted and analyzed using bioinformatics methods, including general physical and chemical properties, hydrophobicity, transmembrane region, signal peptide, conserved domains, secondary and tertiary structure, localization sites in cells, and the phylogenetic tree. Results showed that the candidate coding genes Ss-Ppt1, Ss-Ppt2 and Ss-Ppt3 of PPTase in S. sclerotiorum were selected in the genome. The proteins encoded by the three genes were hydrophilic and nonsecreting proteins. There were lots of alpha helix and randon coil in the secondary and tertiary structure of the three proteins. Additionally, all of the three proteins had either SCOP or ACPS domains, or both. So we speculated that Ss-Ppt1, Ss-Ppt2 and Ss-Ppt3 might be the PPTases of S. sclerotiorum. This work could provide a cue for further study on the function of the PPTase in S. sclerotiorum.
