以319头荷斯坦奶牛为材料,采用PCR-SSCP方法检测牛群HSP70-A8基因的多态性,并对其与DHI测定性能进行关联分析。结果表明,在荷斯坦奶牛HSP70-A8基因的第1外显子1522 bp处发生G>C突变,检测到AA、AB和BB 3种基因型, 其中AA基因型频率为38.56%,AB基因型频率为31.03%,BB基因型频率为30.41%;多态信息含量(PIC)为0.3733,在该位点均处于中度多态;卡方检验表明该荷斯坦奶牛群体处于哈迪-温伯格非平衡状态;群体HSP70-A8的第2至8外显子都没有检测到多态。关联分析结果表明,BB基因型荷斯坦奶牛个体总产奶量、标准乳总产奶量、305 d标准乳产量、305 d乳脂产量、305 d乳蛋白产量、总乳脂产量和总乳蛋白产量皆极显著高于AA型个体和AB型个体(P<0.01),AA基因型个体体细胞数极显著低于AB型个体和BB型个体(P<0.01)。因此,HSP70-A8基因第1外显子该位点的多态性可以作为奶牛生产性能选择的候选分子遗传标记。 |

Single nucleotide polymorphism detection of the HSP70-A8 gene by PCR-SSCP and analysis of its relationships with DHI performance measures in Holstein cattle |

319 Holstein cattle were used to detect the polymorphism of the HSP70-A8 gene using PCR-SSCP method. The results showed that a G>C mutation occurred in 1522 base position in exon 1 of HSP70-A8, and three genotypes (AA,AB and BB) were found, in which genotypic frequency of AA was 38.56%, AB was 31.03% and BB was 30.41%. The polymorphism information content (PIC) was 0.3733 in the population, showing a moderate polymorphism. According to the results of χ2 test, the locus was in Hardy-Weinberg non-equilibrium state in the population. No pdymorphism was detected in exon 2 to 8 of the HSP70-A8 gene in the population. It was investigated that cows with BB genotype had higher lactation to date milk yield, lactation to date fat-corrected milk yield, 305-day fat-corrected milk yield, 305-day fat yield, 305-day protein yield, lactation to date fate yield and lactation to date protein yield than cows with AA and AB genotypes (P<0.01). Cows with AA genotype had lower somatic cell numbers than cows with AB and BB genotypes (P<0.01). It was suggested that exon 1 of HSP70-A8 should be selected as one of indicator molecular marker genes for milk performance of Holstein cattle. |
