GPR54基因C-816T/T-754C品种间分布 及其与早熟性的关联

摘要:
GPR54基因是牛早熟性状的重要候选基因。以42头本地黄牛、44头西门塔尔牛及其116头杂交育种群等为试验群体,分析牛GPR54基因5’调控区C-816T/T-754C在不同牛群体中的分布、启动子效率及其与牛早熟性状的关联性。提取牛耳组织或血液基因组后,通过PCR产物测序鉴别SNP位点,双荧光素酶报告基因验证其SNP位点对启动子效率的影响。结果表明,在GPR54基因CDS上游5’调控区检测到T-754C和C-816T 2个SNP位点;西门塔尔牛和安徽地方黄牛分别只有1种基因型,即CCTT和TTCC,单倍型存在连锁不平衡。报告基因验证2种单倍型的启动子有明显差异,其中-816CT-754启动子的效率要比-816TC-754提高34.31%(P<0.01)。关联分析显示,816CC754TT基因型牛的初情期比816TT754CC基因型牛的初情期提前了1.28月。

Distribution of C-816T/T-754C locus in the GPR54 gene in different cattle breeds and their association with precocity

Abstract:
The GPR54 gene is an important candidate gene for bovine early maturity characters. Forty-two Anhui local cattles (AL), 44 Simmental cattles (ST), and 116 hybrid cattles (HC) from the breeding group of AL×ST were selected. The distribution of C-816T/T-754C locus in GPR54 and their association with the precocity trait were analyzed in different cattle breeds. All DNA samples were extracted from the ear tissue or blood. SNP loci in target fragments were identified by sequencing the PCR products. The effects of the SNP loci on the promoter efficiency were validated using a dual luciferase report gene. The results showed that two SNP loci, T-754C and C-816T, in 5’ regulatory region of the GPR54 gene were detected. The genotypes of ST and AL were CCTT and TTCC, respectively, with linkage disequilibrium using the haplotype analysis. The efficiency difference between two haplotype promoters was significant in which the efficiency of -816CT-754 promoter increased 34.31% than that of -816TC-754 promoter (P<0.01). A correlation analysis showed that cattle puberty of the 816CC754TT genotype was 1.28 months earlier than that of the 816TT754CC genotype (P<0.01).
