CD-1小鼠孕晚期暴露LPS对中老年期焦虑和探索行为的影响

摘要:
检测CD-1小鼠孕晚期暴露细菌脂多糖(LPS)对其中老年期焦虑和自发性探索活性行为的影响。结果显示,15月龄CD-1小鼠在3个任务中的焦虑水平均较青年组升高,自发性探索活动降低(Ps< 0.05)。接受LPS处理的15月龄小鼠与同龄对照组比较,高(50 μg·kg-1)和低(25 μg·kg-1)剂量组在旷场中的周边时间均显著长,跨格子数显著少(Ps< 0.05),高剂量组的直立次数也减少(P< 0.05)。在黑白巷中,高剂量组的潜伏期和黑巷时间均显著长于同龄对照组(Ps< 0.05)。高剂量组在高架十字迷宫中的直立次数显著少于同龄对照组。上述结果提示CD-1小鼠孕晚期暴露LPS可剂量依赖地加速其中老年期任务特异性焦虑和自发性探索活动行为改变。

Effects of lipopolysaccharide exposure during the late pregnancy on anxiety and locomotor behaviors of the middle-aged CD-1 female mice

Abstract:
The aim of the present study was to determine whether system inflammation during the late gestation period affects anxiety and locomotor behaviors of the middle-aged female CD-1 mice. At day 15 to 17 in the gestation peroid, the experimental mice received daily intraperitoneal injection of normal saline (NS), 50 μg·kg-1 LPS (high dose, HD), or 25 μg·kg-1 LPS (lower dose, LD). An anxiety-based test battery was completed at the age of 15 months with three behavioral tasks including open field, black-white alley, and elevated plus maze. A group of 5-month old mice was the young control. Results showed that the NS group showed the increased anxiety and decreased locomotor activity compared to the young control as measured by the three tasks (Ps<0.05). The HD group had significantly more peripheral time and fewer squares crossed and rears than the NS group in the open field (Ps<0.05). The LD group also had fewer squares crossed and rears than the control (Ps<0.05). In the black-white alley, the HD group had a significantly increased latency and time in the black alley than the NS group (Ps<0.05). The HD group had fewer rears than the NS group in the elevated plus maze (P<0.05). The results suggested that exposure to lipopolysaccharide during pregnancy could induce a dose-dependent deterioration of an anxiety increase and locomotor decline in the middle-aged female CD-1mice.
