一种低浓度L-半胱氨酸介导的表没食子儿茶素 没食子酸酯红色衍生物的制备方法

摘要:
优化低浓度L-半胱氨酸介导的表没食子儿茶素没食子酸酯的红色衍生物的制备条件。其最佳反应条件为,在pH6.0磷酸-柠檬酸缓冲溶液中,80℃水浴反应3 h,底物表没食子儿茶素没食子酸酯浓度为0.15 mg·mL-1,L-半胱氨酸浓度为0.01 mg·mL-1。并探究该条件下制备的红色衍生物的固体粉末的外观特性、溶解特性、光谱学特性以及HPLC色谱行为,为后续的分离纯化、结构鉴定打下基础。

Abstract:
This study optimized the preparation condition for low concentration L-cysteine mediated epigallocatechin gallate (EGCG) red derivatives. The reaction condition was optimized and established as: incubation in pH 6.0 phosphate - citric acid buffer solution for 3 hours at 80℃ with substrate 0.15 mg/ml EGCG and 0.01 mg/ml L-cysteine. Besides, we investigated the appearance characteristics, solubility, spectroscopy and HPLC chromatographic behavior of the red derivatives solid powder under the preparation condition, which would lay a solid foundation for further separation and structure purification of the red derivatives.
