蝴蝶兰“V31”品种花梗腋芽快繁体系的建立

摘要:
以蝴蝶兰“V31”品种花梗腋芽为试验材料,通过筛选丛生芽诱导、增殖、生根及移栽各阶段最适培养基,建立其快繁体系。结果表明,最佳腋芽诱导丛生芽培养基MS + 8.0 mg·L-1 6-BA +2.0 mg·L-1 NAA + 15 g·L-1椰子粉,诱导率84.57%;最佳丛生芽增殖培养基MS +10 mg·L-1 KT +0.6 mg·L-1 NAA + 0.2 mg·L-1 2,4-D,增殖倍率7.94;最佳生根壮苗培养基1/2 MS +0.1 mg·L-1 NAA + 1 mg·L-1 6-BA+ 20 g·L-1蔗糖+ 10 g·L-1香蕉粉,平均生根数9.47;最佳栽培基质为经过0.1%高锰酸钾浸泡0.5 h的水苔+玉米棒(比例为1:1)。

Establishment of rapid propagation technique system using pedicel axillary buds of Phalaenopsis

Abstract:
By using pedicel?axillary?buds?of Phalaenopsis cultivar Dtps. Tailin Red Angel“V31” as tissue culture material, the rapid propagation technology system of Phalaenopsis was established, and the optimum mediums for pedicel?axillary buds induction, multiplication, root, and transition were selected, respectively. The results showed that the optimum induction, multiplication, root mediums for Phalaenopsis “V31” were MS+8.0 mg·L-1 6-BA+2.0 mg·L-1 NAA +15 g·L-1 coconut powder, MS+10 mg·L-1 KT+0.6 mg·L-1 NAA+0.2 mg·L-1 2, 4-D, 1/2 MS+0.1 mg·L-1 NAA+1 mg·L-1 6-BA+20 g·L-1 agar+10 g·L-1 banana flour, respectively; the induction and multiplication ratios were 84.57% and 7.94 times, respectively, and the average rooting number per shoot was 9.47. The optimum growing medium was sphagna + cob (with the ratio of 1:1, dipping sphagna and cobs in 0.1% KMnO4 for 0.5 h ).
