鸡气管黏膜上皮基底细胞的分选与鉴定研究

摘要:
鸡气管黏膜上皮细胞是IBV和其他禽病原体感染的重要靶细胞。气管黏膜上皮细胞种类多样,不同细胞对病原感染敏感性有差异,研究与比较IBV感染不同种类气管黏膜上皮细胞特性有助于弄清IBV分子感染机制。利用流式细胞仪技术,使用基底细胞特异性抗体从鸡气管黏膜上皮细胞中分选出基底细胞。通过基底细胞培养形态的观察、生长曲线的绘制和特异性免疫荧光的鉴定,成功获得能够稳定生长繁殖的高纯度的鸡气管黏膜基底细胞,为今后IBV或其他相关研究奠定技术基础和提供了良好的种子细胞。

Sorting and identification of basal cells of chicken tracheal mucosa

Abstract:
Chicken tracheal epithelial cells are important target cells by infected with IBV or other poultry pathogens. Tracheal epithelial cells include different types of cells, each of which has different susceptibility to pathogenic infection. Researching and comparing infection characters of different types of tracheal epithelial cells by IBV helps to clarify IBV molecular infection mechanism. In this study, by using flow cytometry technology and basal cell specific antibody, chicken basal cells of chicken tracheal mucosa were sorted out; by observing the morphology of cultured basal cell, mapping growth curves and specific immunofluorescence identification, high purity chicken tracheal mucosa basal cells with stable growth and reproduction were successfully obtained, which lays a technical foundation for future IBV or other relevant research and providing good seed cells.
