早期断奶对仔猪肠上皮细胞氧化与损伤的影响

摘要:
为了研究早期断奶对仔猪肠上皮细胞氧化与损伤的影响,将80头21日龄健康哺乳仔猪随机分成2组,吮乳组与断奶组,分别于断奶0、1、3、5、7、10和14 d采样检测各组仔猪肠上皮细胞中氧化、损伤、修复相关指标。结果显示,与吮乳组相比,断奶组肠上皮细胞中抗氧化酶活性显著降低、过氧化产物丙二醛(MDA)含量明显升高,二胺氧化酶含量先降低后升高、肠上皮细胞损伤明显,表皮生长因子及其受体的mRNA表达量迅速降低,并显著低于吮乳组, 氧化还原因子的mRNA表达显著降低。综合本试验指标所得数据结果表明,断奶后1~3 d急剧变化,5~7 d达到峰顶或峰谷,而后开始恢复,10~14 d恢复到正常水平。这说明早期断奶对仔猪肠上皮的影响持续时间较长,可引起仔猪肠上皮细胞严重的过氧化,损伤严重,自然修复较缓慢。

Effects of early-weaning on intestinal antioxidation and impairment in the piglet

Abstract:
The purpose of this research was to evaluate the effects of early weaning on intestinal epithelial cell antioxidation and impairment in piglets. Eighty piglets aged 21 days were randomly allocated into suckling and weaning groups. Piglets were scarified in each group on 0, 1, 3, 5, 7, 10 and 14 days after weaning to examine the indexes about oxidation, impairment and recovery. As a result, compared with the suckling group, the activities of antioxidational enzymes increased significantly in intestinal epithelial cells in the weaning group. The concentration of MDA was significantly higher in weaning group than that in the control. In addition, the activity of diamineoxidase (DAO) was reduced at first, and then rose (P<0.05); the impairment of intestinal epithelial cell was serious; the expression of the EGF, EGFR and Ref-1 was declined in some degree and lower in weaning group than that in the control (P<0.05). Taken together, all the indexes changed sharply during days 1 to 3 of weaning, reached to the highest or lowest during days 5 to 7 of weaning, and recovered to normal level during days 10 to14 of weaning. It clarified that early weaning could cause long duration to piglets’ intestinal epithelial cells and peroxidation to intestinal epithelial cell. The intestinal epithelial cell was severely impaired, and its recovery was very slow.
