鸡源大肠埃希菌质粒介导的喹诺酮耐药基因的检测与分析

摘要:
为了解安徽省合肥地区鸡源致病性大肠埃希菌对氟喹诺酮类药物(fluoroquinolones, FQs)的耐药性及质粒介导的喹诺酮耐药基因(plasmid-mediated quinolone resistance, PMQR)的流行情况,对从合肥地区多个规模化养鸡场病鸡病料中分离保存的37株疑似大肠埃希菌进行细菌分离培养、生化编码鉴定和致病性测定。利用K-B纸片琼脂扩散法检测鸡源致病性大肠埃希菌对4种FQs的耐药性, 设计并合成4对特异性引物通过菌液PCR法对所分离细菌进行PMQR基因(qnrA、qnrB、 qnrS、 qepA)扩增。结果显示,所检测的37份疑似病料均为致病性大肠埃希菌;37株大肠埃希菌中对FQs 呈3耐以上(包括3耐)菌株占67.57%(25/37);37株大肠埃希菌中检测到qnrA基因,检出率为56.77% (21/37),而qnrB、qnrS、qepA基因均未检出。提示合肥地区鸡源致病性大肠埃希菌对FQs耐药性严重,PMQR基因以qnrA基因为主,且qnrA基因的流行与FQs耐药性具有相关性。

Detection and analysis of plasmid-mediated quinolone resistance genes of Escherichia coli isolates from chickens

Abstract:
To investigate the fluoroquinolones (FQs) resistance of Escherichia coli isolates from chickens in Hefei area of Anhui province and the prevalence of plasmid-mediated quinolone resistance(PMQR)genes, 37 Escherichia coli strains were identified by bacteria cultivation, biochemical code identification and pathogenic determination. K-B diffusion method was adopted to detect FQs resistance of Escherichia coli , and PMQR genes(qnrA, qnrB, qnrS, qepA)were detected by PCR amplification. The results showed that 37 suspected Escherichia coli strains were identified as pathogenic Escherichia coli. The resistance rate to above 3 (include 3) antimicrobials of FQs was 67.57% (25/37). The prevalent rate of qnrA gene was 56.77% (21/37), and qnrB, qnrS, qepA genes were not detected. The results suggested FQs resistance of Escherichia coli strains from chickens in Hefei area is rather serious, PMQR genes focuses on qnrA gene, and the prevalence of qnrA gene is correlated with the FQs resistance.
