培养条件对串珠镰刀菌提取物抑制蛋白核小球藻的影响

摘要:
以对蛋白核小球藻生长抑制率和提取物质量为评价指标,采用正交设计方法,研究了培养条件对串珠镰刀菌(Fusarium moniliforme)提取物及其抑制蛋白核小球藻(Chlorella pyrenoidosa)活性的影响。结果表明,培养温度、碳源、氮源、无机盐组合、pH值、接种量等因子对串珠镰刀菌提取物及其抑藻活性具有显著影响。优化的串珠镰刀菌培养条件为:培养温度22℃,培养液组成为麦芽糖(2%)、蛋白胨(2%)和CaCl2+ MgSO4(0.1%+0.05%),pH 8、接种量为每瓶13个菌饼、装液量每250 mL为160 mL、振速为140 r·min-1。优化条件下,串珠镰刀菌提取物对蛋白核小球藻的EC50为0.33 g·L-1。本试验结果为串珠镰刀菌次生代谢物中抑藻活性物质的分离、鉴定,进而开发新型抑藻剂提供了依据。

Effects of incubation condition of Fusarium moniliforme on anti-algal activity of Fusarium moniliforme extracts

Abstract:
The effects of culture conditions on Fusarium moniliforme extraction and its inhibitor of Chlorella pyrenoidosa growth were studied through orthogonal experiments, taken anti-algal rate and extraction’s mass as indices. The results were as follows. The optimum culture conditions were 22℃, 2%malt sugar, 2% protein peptone, CaCl2+MgSO4(0.1%+0.05%), pH 8, 13 fungus cakes per bottle for the vaccination quantity, 160 mL per 250 mL for nutrient solution and 140 r·min-1 for vibration velocity . Every factor of culture conditions also affected anti-algal rate and extraction’s mass significantly. The EC50 on Chlorella pyrenoidosa by anti-algal compounds is 0.33 g·L-1 under optimum culture conditions. The experimental results provide a basis for separating and appraising secondary metabolite of Fusarium moniliforme, and then for developing a new preparation on algae.
