冬虫夏草子囊孢子及其无性型在培养过程中的形态学观察

摘要:
在显微镜下挑取冬虫夏草弹射的单子囊孢子接种于平板上,18℃培养,在显微镜下观察其生长过程,发现单子囊孢子产生芽管后,直接进行微循环产孢,产生被粘液包裹的球状分生孢子团,经过缓慢生长形成菌落后,开始无性繁殖产生大量分生孢子。取其分生孢子接种到液体培养基中,在18℃下振荡培养,观察其生长过程。分生孢子液态培养主要经过:(I)分生孢子萌发进行微循环产孢;(II)孢子萌发产生菌丝;(III)菌丝产生芽生孢子;(IV)芽生孢子生长形成新的芽孢。结果表明,分离得到的冬虫夏草的无性型是中国被毛孢,且分生孢子在液体培养中也存在微循环产孢的现象。

Morphological observation of ascospores of Ophiocordyceps sinensis and its anamorph in growth process

Abstract:
The single ascospore of Ophiocordyceps sinensis, isolated by the method of the single spore isolation, was cultured in the plate at 18℃. The growth process of the single ascospore was observed by microscope. After the generation of germ tube, spherical spore mass covered by mucus layer was generated through microcycle conidiation of ascospores. The single ascospore developed into colony slowly and then generated a large number of conidia. The conidia were inoculated into the liquid medium at 18℃ in shaking incubator. The culture broth was sampled at certain time and the morphologies of the conidia were observed by microscope as well. The result showed that the conidia generated by the process of microcycle conidiation of ascospores produced a large number of conidia. Then the conidia grew into hyphae. Blastospores emerged from the hyphae and at last blastospores reproduced the new blastospores. The result indicated that the isolate is Hirsutella sinensis, anamorph of Ophiocordyceps sinensis, and its conidia also had the phenomenon of microcycle conidiation in liquid culture media.
