我国部分地方鸡种COⅠ基因多态性及其分子系统进化研究

摘要:
以我国7个地方鸡种为研究对象,利用DNA测序技术测定线粒体细胞色素C氧化酶亚基Ⅰ (cytochrome c oxidase Ⅰ,COⅠ) 基因序列,分析地方鸡种COⅠ多态性及其分子系统进化关系。研究结果表明,选择的这段COⅠ基因序列有24个突变位点,为13个单倍型,其中11个单倍型为各品种所特有,可以作为各品种鉴定的依据;各鸡种在不同位点突变频率不同,7个鸡种均有其特异位点,根据这些特异位点,可以对其进行品种鉴定。7个品种间Kimura 双参数遗传距离为0.001 81~0.008 48。7个品种的DNA分类和形态学分类基本一致,该基因可以用于探讨地方鸡种分类问题。

Genetic diversity and phylogenetic analysis of COⅠgene in some indigenous chicken breeds

Abstract:
The COⅠgene in seven indigenous chicken breeds was amplified using PCR and sequencing. The sequences were analyzed to explore utility as DNA Barcoding for identifying these chicken breeds. The study revealed that there were 24 polymophic sites and 13 haplotypes in the COⅠgene, of which, 11 were special to seven chicken breeds. The special sites were found in each chicken breed which was the bases to identify chicken breeds. Kimura 2-parameter distance between the 7 chicken breeds was 0.001 81-0.008 48. DNA taxonomy is consistent with morphological taxonomy among the seven chicken breeds. The phylogenetic relationships of the breeds can be well resolved by COⅠ gene sequences.
