牛精子蛋白质组的双向电泳和质谱鉴定初步研究

摘要:
通过建立牛精子双向电泳-质谱鉴定研究平台,为从蛋白质水平研究精子生理机能奠定技术基础;采用改进的热TRIzol法裂解精子细胞制备蛋白,应用双向凝胶电泳(2-DE)分离牛精子蛋白,通过基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定蛋白质点。得到了重复性较好的牛精子2-DE图谱,质谱鉴定16个蛋白质点。建立牛精子双向电泳-质谱鉴定研究平台,为后续牛精子X、Y差异蛋白点的检测和分析奠定了理论和技术基础。

Abstract:
In order to lay a technological foundation for studying physiological function of spermatozoa,proteomics research of bovine spermatozoa with two-dimensional gel electrophesis and MALDI-TOF-MS were established.The heat TRIzol method and two-dimensional gel electrophoresis(2-DE) were used to separate the total proteins of bovine X and Y spermatozoa,ImageMaster 6.0 software was applied to analyze 2-DE images,and proteins were identified by both MALDI-TOF-MS and 2-DE.As a result,16 proteins in the spermatozoa w...
