瓜蒌炭疽菌培养基优化研究

摘要:
采用二次通用旋转组合设计方法对2个瓜蒌炭疽菌菌株的培养基的最佳组合进行研究,建立具有较好预测性能的培养基反应模型,并验证了该模型的可靠性;利用该模型探讨培养基最优组合的各种重要单因子对菌丝生长量反应规律及交互作用.结果表明,S1与S9菌株对生长因子的要求不同,3个因子对S1与S9菌落直径影响大小顺序为pH>葡萄糖>组氨酸;2个因子交互作用对S1菌落直径影响的大小顺序为葡萄糖与组氨酸互作 > pH与葡萄糖互作> pH与组氨酸互作,对S9菌落直径影响的大小顺序为葡萄糖与组氨酸互作 > pH与组氨酸互作>pH与葡萄糖互作.

Abstract:
The optimization of medium on the two isolates of anthracnose in Trichosanthes kirilowii was studied by using the general quadratic rotatory regression of three factors and five levels in this experiment.The effects of combined pH value, glucose content and histidine content on the colony diameter on PDA plate and mycelium dry weight in Czapek liquid medium were studied systemically. The mathematic model of reaction on the medium was built up, which was of high predication and verified to be reliable by the experimental data.The reaction regularity and relation between each factor in the best combination of medium and the mycelium growth value and the mutual effect of different factors were discussed by use of the mathematic model, and the factors which affected the growth of two isolates was also compared.The results showed the requirements were different between the growth of S1 and that of S9.The order of the effects of the three factors on the two isolates colony diameter was pH value>glucose content>histidine content. The order of the effect of the interaction between two factors on the S1 colony diameter was the interaction between glucose and histidine>the interaction between pH and histidine>the interaction between pH and glucose.
