Abstract:The new early-maturing potato varieties ‘Wanmalingshu 1’ and ‘Wanmalingshu 2’ were bred and selected by Institute of Horticulture, Anhui Academy of Agricultural Sciences. Due to the genetic background difference, in vitro plantlets cultured on traditional Murashige & Skoog (MS) medium were weak and could not meet further cultivation requirements. In order to obtain a suitable culture medium, in vitro potato plantlets of ‘Wanmalingshu 1’ and ‘Wanmalingshu 2’ were inoculated on MS medium supplemented with different concentrations of 1-Naphthylacetic acid (NAA) and 6-Benzylaminopurine (6-BA) to understand plant growth regulator effect on production of robust in vitro plantlets. The results showed that, the optimum medium for ‘Wanmalingshu 1’ is MS + 0.5 mg·L-1 NAA + 0.1 mg·L-1 6-BA, and the suitable medium for ‘Wanmalingshu 2’ is MS + 1.0 mg·L-1 NAA + 0.1 mg·L-1 6-BA.
