Abstract:The fresh algae mud from Chaohu was used to obtain the crude extract of phycocyanin (PC) using freezing and thawing. The crude extract was purified by two-step salting-out, and the PC was purified by cellulose (Cellufine A-500) anion exchange resin and finally purified by hydroxyapatite (HA) column. The results showed that At room temperature (25℃), the optimum molar concentrations of two-step salting-out (NH4)2SO4 were 1.0 mol·L-1 and 1.8 mol·L-1, respectively. The PC purity (A620 / A280) reached to 2.168 and the PC recovery was 37.5 %. The purity of PC (A620 / A280) reached 3.211 and 4.133, respectively, and the recovery was 20.3% and 7.2%, respectively, which reaches the reagent grade purity.
