    通过对云龙矮脚鸡线粒体DNA(mtDNA) D-loop区全序列的测序和比对,并结合GenBank已经测出D-loop区全序列的红色原鸡的序列,探讨云龙矮脚鸡的遗传多样性和母系起源。结果显示,云龙矮脚鸡mtDNA D-loop区全长1231~1232 bp,1231 bp单倍型在859 bp处存在C碱基缺失。在19只个体中,共发现25处单核苷酸多态位点,8种单倍型。系统发育分析发现,云龙矮脚鸡8种单倍型可以分为A、B、C和E 4个分支。A和B与红色原鸡滇南亚种(Gallus gallus spadiceus)聚为一类,推测这2个分支可能起源于云南、缅甸的附近地区的红色原鸡滇南亚种。E分支与红色原鸡(Gallus gallus murghi)聚为一类,推测可能起源于红色原鸡的印度亚种分支。D分支与4个原鸡亚种聚为一类,可能有多个母系起源。本研究从分子水平上为云龙矮脚鸡的遗传资源保护和开发利用提供了参考。

    Abstract:

    To determine the origin and genetic diversity of Yunlong Aijiao chicken, we compared its complete mtDNA D-loop sequences with the red jungle fowl sequences in GenBank. The results showed that the D-loop region of Yunlong Aijiao chicken was 1231-1232 bp in length and the 1231 bp haplotype had a nucleotide C deficiency at 859 bp site. In 19 samples, 25 mutation sites and 8 haplotypes were found. Phylogenetic analysis found that 8 haplotypes of Yunlong Aijiao chicken can be classified to four clades. Clade A and clade B were clustered with Gallus gallus spadiceus, which indicated that the two clades might originate from the continental subspecies of Gallus gallus spadiceus. The clade E was clustered with Gallus gallus murghi, indicating that clade E might originate from the continental subspecies of Gallus gallus murghi. Clade D was clustered with four subspecies of the red jungle fowl, which may have multiple origins. The results will be a reference for genetic resources conservation, development, and utilization of Yunlong Aijiao chicken.
