Abstract:In order to verify the correlation between the M2032 locus of Mx gene and avian leukosis in Wuhua chickens, PCR-RFLP method was used to analyze the polymorphism of the Mx gene at 2032 locus. The frequencies of allele A and G at that locus in the experimental group were 3.03% and 96.97%, respectively. Two genotypes, AG and GG, were detected in 99 Wuhua chickens, with the frequencies of 6.06% and 93.94%, respectively. Chi-square test showed that the frequencies of the Mx gene were significantly deviated from Hardy-Weinberg equilibrium. The correlation between genotypes and avian leukosis indicated that the positive rate of avian leukosis in group of AG genotype (16.67%) was numerically lower than that of GG genotype (58.06%). Accordingly, the number of AG genotype would be larger than that of GG genotype. Forty one-day old Wuhua chickens negatively in avian leukosis were used to detect the polymorphism of the Mx gene using the same method described as above; however, frequencies of genotypes AA, AG, GG were 2.50%, 5.00% and 92.50%, respectively, which was inconsistent with the above conclusion. We speculate that the GG genotype of Wuhua chickens might be susceptible to avian leukemia. The positive rate of avian leukosis may vary with age.The susceptibility of avian leucosis of AG genotype would be much lower than that of GG genotype.
