    为研究安徽地方枣种质枣疯病16S rDNA序列特性,以健康及表现枣疯病症状的繁昌长枣叶片DNA为模板,克隆枣疯病植原体16S rDNA序列,获得1条1 248 bp的片段,命名为JWB-FJP1。同源性分析结果表明,JWB-FJP1属于16Sr V-B组,与重庆地方枣枣疯病植原体JWB-Xch(JQ675716)同源性最高,达到99.92%;与河北枣枣疯病植原体JWB-Hebei(GU184186)同源性最低,为67.06%,说明不同地区枣疯病植原体存在一定的生物学特异性。本研究结果为安徽地方枣种质枣疯病病原分类及其快速鉴定提供了理论依据。

    Abstract:

    To study the sequence characteristic of 16S rDNA from jujube witches′ broom in Anhui Province, we extracted the DNA from healthy and infected leaves from Fanchangchang jujube, cloned 16S rDNA sequence of jujube witches’ broom phytoplasma, and gained a fragment of JWB-FJP1 with 1 248 bp in length. The sequence homology analysis showed that JWB-FJP1 shared the highest identity of 99.92% with Chongqing jujube witches’ broom phytoplasma strain JWB-Xch (JQ675716) and shared the lowest identity of 67.06% with Hebei jujube witches’ broom phytoplasma strain JWB-Hebei(GU184186), and it was classified as a member of 16Sr V-B group. The result indicated that phytoplasma showed different biological characteristic in different areas. This research provided theoretical basis for fast identification and classification of jujube witches’ broom phytoplasma in Anhui local jujube species.
