    60Co-γ射线辐射小麦品种河科2号的干种子,对M4代234个株系的蛋白质含量、湿面筋含量、沉降值、硬度的变异进行了分析。以超过群体均值±2×标准差为选择标准,筛选出6个蛋白质和湿面筋含量正向变异株系和3个负向变异株系。在M5代对它们的粉质参数进行了测定,并利用SDS - PAGE对其高分子量麦谷蛋白亚基(HMW-GS)进行了鉴定。结果表明,株系242的粉质图谱与粉质参数显著改变,株系240、108的HMW-GS组成也存在明显差异。亲本的HMW-GS组成为2+7+8+9+12,株系240、108的HMW-GS组成为2+7+8+12,说明在麦谷蛋白位点上发生了变异。

    Abstract:

    Dry seeds of wheat HEKE2 were radiated by 60Co γ-ray with the dose of 300 Gy, and the genetic variation of protein content, gluten content, zeleny and hardness of 234 lines were analyzed in M4 population. The contents of protein and gluten in 6 positive and 3 negative mutant lines were identified based on the selection criterion of over mean±2s. Further, their quality traits and farinogram parameters were detected, and high molecular weight glutenins(HMW-GS)were identified by SDS - PAGE in M5 generation. The result showed that the farinograph map and farinogram parameters of the mutant line 242 changed significantly, and HMW-GS composition of the parent was 2+7+8+9+12, while it was 2+7+8+9 for the mutant line 108 and 240. The obvious change of HMW-GS composition suggests that mutations of glutenin locus may be occurred in the mutant lines.
